I submitted university english language proof for my Spouse but her degree is not complete yet.

I got PCC and Medical email from CO, but he did not ask for IELTS for my wife. Does it mean he has accepted that english language letter or should my wife give IELTS as well to avoid the risk?

Even a Senior Secondary School (12th Grade) certificate mentioning that the language of instruction was English would suffice.
Generally the CO would send a consolidated list of documents needed. So if you have been asked for just PCC and Medical it is 90% chance he does not need anything else.
